Summary:Abstract : <br /> <br /> <br /> The in-vitro and in-vivo effect of penetration enhancers 10% ethanol, 2 and 5% dimethyl sulfoxide, 5 and 10% ethyl acetate, a mixture of 5% ethanol and 10% ethyl acetate on transdermal absorption of diclofenac diethylammonium in hydroxypropyl methylcellulose aqueous gel had been studied. The in-vitro penetration was determined using diffusion cell with Spangler fluid impregnated Whatman paper no. 1 and a phosphate buffer of pH 7,4 as reseptor medium. Quantitation of diclofenac diethylammonium in the receptor medium was done using ultraviolet-visible spectrophotometry. The in-vivo penetration study was carried out using patch test on six New Zealand strain rabbits. Serum diclofenac level was determined by high performance liquid chromatography. The result showed that 10% ethyl acetate enhanced the transdermal diclofenac diethylammonium absorption.
