Methods : HTF’s from glaucomatous eyes divided into 4 groups consist of FBS
2% control group, MMC (Mitomycin C), Fibrin Glue (FG) and LMSCs-CM
treated group. The experiment was conducted in 7 days. Col-1, MMP-3, and
TIMP-2 expression were assessed with immunofluorescence staining. Intensity of
fluorescence was measured using ImageJ software. Difference of expression level
among groups were analyzed using Oneway ANOVA test followed by post-hoc
test with p<0.05 considered to be significant
Results : Col-1 expression were decreased FG (0.78 ± 0.09 pixels) and LMSCs-
CM conditioned media (0.78 ± 0.10 pixels) compared to the FBS 2% control
group (1.52 ± 0.33 pixels) (p = 0,000). However, MMC provides better antibrotic
effects on HTFs revealed with decreased of Col-1 expresion in MMC group (0.41
± 0.28 pixels). MMP-3 expression levels were found to be increased in the MMC
group (1.87 ± 0.58 pixels), FG (1.67 ± 0.48 pixels), and LMSCs-CM (1.58 ± 0.56
pixels) compared to the FBS 2% control group (0.90 ± 0.11 pixels) (p = 0.002). A
decrease in the expression of TIMP-2 showed significant results in the MMC
group (0.72 ± 0.31 pixel), FG (0.73 ± 0.40 pixel) and LMSCs-CM (0.80 ± 0.23
pixel) compared to the PBS control group of 2% (1.36 ± 0.36 pixels) (p = 0.002).
Conclusion : FG and and LMSCs-CM might have antifibrotic effect on HTFs
through modulation of extracellular matrix |
