Summary: | Background: Reseach on the effects of red fruit has been conducted with various result. In this study, the effect of red fruit extract (EBM) on the level of cytokines TNF-α, IL-10 and the parasitemia in swiss mice infected with Plasmodium berghei well beassessed.
Methods: The study used a laboratory quasy-experimental design with pre and post test only control group. Sixty male Swiss mice of 8 weeks old and weighs 20-30 g, was simply randomized into four treatment groups. Group I (K1) was stimulated with EBM for 2 weeks before and 2 weeks after infection with P.berghei. K2 was stimulated with EBM for 2 weeks before infection, K3 was stimulated with EBM for 2 weeks after infection and K4 is negative control was given 0.6% tween 40. The dose of EBM was 7.8 mg/30 g BW mice / po once a day. Serum level of TNF-α and IL-10, was measured by ELISA Sandwich methods and the number of parasitemia were examined microscopically. The differences levels of TNF-α, IL-10 and parasitemia of each treatment group were analyzed by t-test, one way anova, honestly significant different (HSD) and also multivariate analysis (manova).
Results : There are significances differences number of parasitemia in treatment group was stimulated with EBM for 2 weeks after infection, if compared with negative control group, for day-6 is 13,735% and day-9 is 1,054%, and also to the treatment group was stimulated with EBM for 2 weeks before infection, which day-6 is 13,141% and day-9 is 32,455%. But negative control group shows, day-6 is 58,180 % and day-9 is 78,506%. The significances differences to the levels of TNF-α, showed also by treatment group was stimulated with EBM for 2 weeks after infection, which at day-3 is 23,269 pg/ml and day-6, is 22,244 pg/ml. But the negative control shows at day-3 is 69,983 pg/ml and day-9 is 94,679pg/ml. While all treatment group shown increase the levels of IL-10, and not significances differences if compared with the negative control. But the levels of IL-10 from treatment group was stimulated with EBM for 2 weeks after infection, has rise sharply, increase progressively by 252,892pg/ml. It is the highest concentration compared with other treatmen group. The increase levels of IL-10 from treatment group was stimulated with EBM for 2 weeks after infection, with the significantly reduces of parasitemia and the levels of TNF-α. But the increased levels of TNF-α is fatal evident, because with the death of mice, such as showed by treatment group was given EBM for 2 weeks before and 2 weeks after infection and treantemnt group was given EBM only 2 weeks before infection.
Conclusion: The extract of red fruit can reduce the levels of TNF-α and the significantly progrees can increase the levels of IL-10. Red fruit extracts can also significantly reduce the number of parasitemia. The effects of EBM is more significant, especially when administered after malaria infection occurred. |
