Muntingia calabura L. leaves contain tannins, flavonoids, and polyphenolics that believed having capability as antibacterial agent. S. mutans is one of bacteria causes dental caries that has glucosyltransferase (GTF) enzyme. GTF is enzyme that capable to catalyze glucan synthesis in the progression of dental caries. The aim of this research is to know the effect of 10% concentration of Muntingia calabura L. leaves extract toward S. mutans GTF enzyme activity. This research used 10% concentration of Muntingia calabura L. leaves extract, Chlorhexidine gluconate 0.12%, and sterile distilled water, as the treatment, positive control, and negative control, respectively. The method of this research consists of three steps: preparation of 10% concentration of Muntingia calabura L. leaves extract, preparation of GTF enzyme from S. mutans supernatant that has been centrifuged, and GTF enzyme activity assays through fructose extensive area analysis by using High Performance Liquid Chromatography (HPLC). Fructose extensive area determined based on time retention from each groups. Fructose concentrations were expressed in percent (%) then converted into μmol/ml fructose that defined as a unit of GTF enzyme activity. The data was analyzed by one-way ANOVA. The results of one-way ANOVA analysis showed p<0.05. It means statistically significance difference in GTF enzyme activity between the groups. The conclusion of this research is 10% concentration of Muntingia calabura L. leaves extract capable to inhibit S. mutans GTF enzyme activity.
