Summary:Background: Diabetes mellitus is a metabolic diseases that characterized by hyperglycemia which result of defects in insulin secretion, insulin action, or both. Data from WHO showed that prevalention of diabetes extremely increase in the some countries included Indonesia. The easy prevention and therapy action is dietary modification with high fibre and Resistant Starch (RS) diet. Physiological characteristic of fibre and RS, and their metabolism on human colon can help glycemic control. Gembili (Dioscorea esculenta) is the one of high fibre and RS food that possible to help glycemic control. Objective: The aim of this study was to assess the effect of gembili powder (Dioscorea esculenta) on glucose level of wistar rats induced by nicotinamide and streptozotocin. Method: Experimental study was used as a design in this study. Twenty five (25) male Wistar rats aged 2 month were used in this study. Rats was devided into five groups. Group one is healthy control group, group two is hiperglycemia control group, and group three-five is treatment groups. Hiperglycemic control groups and treatment groups was induced with Nicotinamide and Streptozotocin before treatment to make hiperglycemia condition. The treatment groups was given gembili powder 1,25 g, 250 g, and 5,00 g each day. Glucose test was doing before treatment, 2 week after treatment and 4 week after treatment. Result: The significant decrease of glucose level was occur on 1,25 g and 2,50 g gembili powder treatment with p value 0,001 and 0,002. Highest decrease glucose level was happen on 2,50 g gembili powder perday treatment. Conclusion: 1,25 gr and 2,50 gr Gembili powder (Dioscorea esculenta) perday treatment could decrease glucose level on Nicotinamide and Streptozotocin Induced Rats
