Summary: | Aphthous stomatitis is an oral wound which can be worsened due to food mastication and talking. Gamavuton-0 (GVT-0) a curcumin analogue which has an anti inflammatory activity. The purposes of this study were to develop and optimize formulations of GVT-0 buccoadhesive film using chitosan and PVP K-30 and evaluate its effect on wound healing process based on numbers of polymorphonuclear cells in the rat buccal mucosa.
Film formulas were determined by using simplex lattice design. Film�s physicochemical evaluations were done to determine the effect of chitosan and PVP K-30. The best obtained formula was used to study the transport of GVT-0, ex vivo residence time and its in-vivo anti inflammatory activity using negative control, placebo matrix control and treatment group. Each group consisted of 3 randomly selected male Wistar rats with the body weight range of 200-300 g which were sacrificed on days 1, 3, 5, and 7 post injury. The injury was made by biopsy punch (d=2.5mm).
All film formulations were thin, flexible, smooth and transparent with weight and thickness uniformity. GVT-0 content in the films ranged from 91-100%. The GVT-0 release data fit well to the Higuchi expression and possesses suitable swelling profile and produce no irritation. The optimum ratio GVT-0 buccoadhesive film is 1.702% chitosan and 1.298% PVP K-30. In-vitro drug release from optimum formula showed that about 45.03±0.22% drug was released up to 6 hours and ex-vivo mucoadhesive time was found to be 125± 3.78 minutes. The GVT-0 permeation through rabbit buccal mucosa was found to be 2.30±0.125 for 6 hours. GVT-0 buccoadhesive film significantly can decrease the number of polymorphonuclear cells (p<0.05). In conclusion, buccoadhesive films of Gamavuton-0 can be proposed as a new therapeutic tool for aphthous stomatitis with drug was released locally and only a very small amount might have absorbed systemically. |
