Summary:Acid Phosphatase was isolated and purified from cassava (Manihot Esculenta Crantz syn M. utilissima Pohl) and Kamote (Ipomea batatas) by ammonium sulfate precipitation, centrifugation, and dialysis. The protein concentration was determined through Biuret's test. The activity of the enzyme was monitored using the conversion of p-nitrophenyl phosphate to p-nitrophenol by acid phosphatase. The colored product of the reaction was determined spectrophotometrically. High Performance Liquid Chromatography was used to confirm the presence of acid phosphatase.
