Glucomorirngin is as a known bioactive compound, which has many excellent properties such as inflammation, stress response, phase I metabolism, antioxidant activities, and direct antimicrobial. In this study, multiple-stage microwave assisted extraction (MAE) of glucosinolates from Moringa oleifera seed meal was investigated to make the extraction of glucosinolates to be done in shorter extraction time and higher yield. The data of single-stage MAE was analyzed by RSM. The effects of three main parameters, extraction temperature (60°C, 70°C, 80°C) solvent-to-solid ratio (20:1 mL/g, 30:1 mL/g, and 40:1 mL/g), and extraction time (5 min, 10 min, and 15 min) were investigated while the subsequent yield was optimized. The glucosinolates yield from the study ranges from 84.291 mg/g to 217.961 mg/g seed meal. The optimal condition for extraction glucosinolates were 37.5 mL/g and 69°C and 5 min of solventto- solid ratio, extraction temperature, and extraction time, respectively. The maximum predicted value of total glucosinolates under optimal conditions were 217.60 mg/g seed meal, which were close to the experimental values was 213.914 mg/g. In this study, using MAE, the yield of glucosinolates was 28.2% higher than using conventional method. Multiple-stage MAE was performed by using the optimal parameters from single-stage MAE. By applying multiple-stage MAE, the yield of glucosinolates was increased from 217.60 mg/g to 239.20 mg/g. This result showed that 90.97%, 7.75%, 1.03%, and 0.17% of glucosinolates were obtained at first, second, third, and fourth extraction stage, respectively. In this study, two stage extraction was a good process to extract almost entirely glucosinolates from Moringa oleifera seed meal.
