Summary:Since its discovery 12 years ago, mortalin/mtHSP70/GRP75/PBP74 has emerged as a heat shock protein at the crossroads of mitochondrial energetic, aging and carcinogenesis. Because of its chaperone nature, depending on sub-cellular localization and on its binding partners, mortalin can assume dynamic and multiple functions relevant to cell survival, control of proliferation, cellular homeostasis and stress response. This protein is now beginning to be appreciated in various other field of biomedical research, including toxicology and immunology. In this chapter, we explore the first dozen years of research on mortalin and expound on its unique promises in biotechnology and biomedicine.
