Identification of biomarker in patients on low dose warfarin : an iTRAQ-coupled LC-MS/MS approach

Warfarin is one of the most widely used oral anticoagulant. However, the dosage of warfarin given to patients is very critical as an overdose may lead to severe bleeding or an under-dosing may lead to clotting of the blood. Hence, it is important to find clues and leads to enable medical professiona...

Chen Wei Ning, William School of Chemical and Biomedical Engineering DRNTU::Engineering::Chemical engineering::Biotechnology Warfarin is one of the most widely used oral anticoagulant. However, the dosage of warfarin given to patients is very critical as an overdose may lead to severe bleeding or an under-dosing may lead to clotting of the blood. Hence, it is important to find clues and leads to enable medical professionals to determine if individual patients are to be on high or low dose warfarin requirements. The search for protein biomarkers which helps to differentiate patients on low-dose warfarin requirement from those on high-dose warfarin requirement is the aim and target of this project. An iTRAQ coupled LC-MS/MS approach will be taken to determine the protein biomarker. From present and past results obtained, TP, a protein biomarker has been found significant in patients on low-dose warfarin and is absent in all high-dose warfarin patients. This observation has led us to determine this protein as our target protein biomarker for patients who are on low-dose warfarin requirement. This finding could potentially minimize the risk of over-dose during warfarin induction.
