Summary: | We aim to engineer a multipurpose M13KE phage display vectors which contains an additional gene III for peptide or protein display to create biosensors for glucose blood sugar levels to help fight diabetes. A truncated gene III was cloned and a fragment containing a lac/tac promoter and the gene III signal peptide sequence was assembled using overlapping-extension polymerase chain reaction. The fragments were joined using PCR via their overlapping regions. Expression analysis with anti-M13 pIII monoclonal antibody revealed their recombinant gene III expression is independent on inducer concentration, but has a temporal variation. |
