This project is focused on the study of mechanotransduction of Human mesenchymal stem cells (hMSC). A mechanotransduction device was custom made to stimulate mechanotransduction to stem cells by stretching, bending or shearing cultured materials. Gelatin immobilized Poly (£-caprolactone) (PCL) was used for mechanotransduction stimulation. The response of hMSC was monitored during the mechanotransduction stimulation while culturing. Upregulation of desmin, MAP2, ALP was observed in single axial stretching condition and Upregulation of NKx2.5 in was observed in bi axial stretching condition. The amplitude of stretching strain was studied by single axial stretching at 1.25%, 2.5%, 5%, 10%, 20% strain. GATA4, NKx2.5, Desmin, Nestin, MAP2 was observed at 1.25% single axial strain stretching. For 2.5% of stretching, gene upregulation of GATA4, desine, MAP2, ALP was observed. It suggests that gene upregulation of hMSC by mechanotransduction is strain dependent.
