Summary:Malignant melanoma accounts for 75% of skin cancer death and the 5-year survival rate is less than 5%, due to the lacking of effective treatment for tumor metastasis. Preliminary study has identified RNF11 as a potential regulator of melanoma metastasis by using insertional mutagenesis. RING finger protein 11 is a 154-amino acid modular protein that is expressed ubiquitously in human. Using overexpression and knockdown of RNF11, we show that RNF11 promotes cell invasion, migration and proliferation of B16F10 murine melanoma cells. We observed up-regulation of Smad-dependent and Smad-independent TGF- β signaling pathways and these pathways may be responsible for the cellular responses of RNF11. We propose that the interaction between Smad-dependent and Smad-independent TGF- β signaling determines the final outcome of cellular responses.
