Summary:Tethered Particle Motion (TPM) is a technique in the biophysical research of bio-polymer mechanical property and bio-molecules structural interaction. In this FYP, a fluorescent three dimensional TPM microscopy is constructed. The designs of optic, channel fabrication and software are discussed in the thesis as well. The developed TPM technique is more accurate, more sensitive and able to provide real time feedback to users. Fundamental experiments are repeated to demonstrate the new system’s stability. Tethered DNA end fluctuation three dimensional tracking is performed to unveil its physiological motion. The system also shows its capability in analyzing fundamental biophysical models and performing future biological researches.
