Summary:The hydrolysate from the action of crude Penicillium oxalicum KUB-SN2-1 mannanase was determined by Thin Layer Chromatography (TLC). The main products that were obtained from copra meal and coffee residue after hydrolyzed by crude mannanase were mannotetraose, mannotriose, mannobiose and mannose while the product of potato peel and sugarcane bagasse was glucose. For soybean meal, the main products were glucose and galactose. Interestingly, the hydrolysate from the degradation of agricultural wastes possessed prebiotic properties in that they promoted growth of 10 strains of Lactic Acid Bacteria (LAB) including Lactobacillus plantarum TISTR 541, Lactobacillus sake TISTR 840, Lactobacillus sake TISTR 890, Lactobacillus sake TISTR 912, Lactobacillus reuteri KUB-AC-5, Lactococcus lactis ATCC 19435, Pediococcus pentosacens JCM 5885, Pediococcus acidilactici TISTR 953, Enterococcus faeclis TISTR 927 and Enterococcus faeclis JCM 5805. In addition, the same hydrolysate could inhibit the growth of pathogenic bacteria including Staphylococcus aureus TISTR 029 and Escherichia coli E010, but not Shigella DMST 1511. The hydrolysate from coffee residue effectively inhibited the growth of Staphylococcus aureus TISTR 029 at the level 76.190 ± 3.88% while the soybean meal hydrolysate moderately inhibited the growth of Staphylococcus aureus TISTR 029 as well as E. coli E010 at the level of 34.78 ± 4.35 % and 20.0 ± 9.0%, respectively. For the aspects of health benefits and value added agricultural wastes, these mannooligosaccharide products are an attractive candidate as an environmental friendly feed supplement.
