A new 3D design of paper-based device on nitrocellulose (NC) membrane with the BCIP/NBT substrate kept on soluble solid for fully automatic multistep sequencing of sandwich ELISA has been developed to improve sensitivity, mass productivity and lifetime of the device. The detection limit of the 3D device with the BCIP/NBT substrate kept on soluble solid was found to be 1.01 mIU/mL which is lower than that with direct immobilization (10.7 mIU/mL). This device can be applied for a greater variety of clinical (point of care assays) or environmental analysis.
