Summary: | a-Amylase, 1,4-a-D-glucan glucanohydrolase (EC 3.2.1.1) is a biocatalyze which hydrolyses a-1,4 glycosidic linkages of starch to produce oligosaccharide and dextrin. a-Amylases from microorganism have been studied but only a few marine organism identified have an a–amylases activities. Kakaban lake is a landlocked marine lake. The purpose of this research was to screen, isolate and characterize of potato degrading a–amylase from Kakaban lake isolates, East Kalimantan. Screening of a–amylase activity from 40 Kakaban Lake isolates by red amylopectine gave 6 isolates showing starch degrading activity. Morphology and physiology analysis of 16S rDNA gene show that isolates KBU 1-6L and KBU 2-FL are Bacillus epiphytus and Pseudoalteromonas sp., respectively. Zimogram analysis also showed that they secrete a-amylases. The activity of isolate KBU 1-6L a-amilase from saturated ammonium sulphate 0-80% fraction at 50 oC, pH 7.0 was 169. 17 U/mL. |
