Kakaban lake is a landlocked marine lake. The purpose of this research was to screen, isolate and characterize of potato degrading a–amylase from Kakaban lake isolates, East Kalimantan. Screening of a–amylase activity from 40 Kakaban Lake isolates by red amylopectine gave 6 isolates showing starch degrading activity. Morphology and physiology analysis of 16S rDNA gene show that isolates KBU 1-6L and KBU 2-FL are Bacillus epiphytus and Pseudoalteromonas sp., respectively. Zimogram analysis also showed that they secrete a-amylases. The activity of isolate KBU 1-6L a-amilase from saturated ammonium sulphate 0-80% fraction at 50 oC, pH 7.0 was 169. 17 U/mL. text
