Summary:Lactic acid bacteria (LAB) is commonly used in food processing and known to produce secondary metabolite showing antimicrobial activity and hence can be developed as biopreservative. The development and utilization of LAB can be conducted by single culture microbe as well as microbial consortium. The aim of this research was to study the activity of bacterial consortium consist of Bifidobacterium, Lactobacillus plantarum 14-BL and Pediococcus acidilactici F11 in producing bacteriosine as antimicrobe using edible growing media made of sweet potato. Consortium ability of the three LAB was conducted by synergetic test. Optimum mixing time of the three LAB and optimum fermentation time to produce bacteriocine-rich supernatant was determined by means of growth curve plotted between turbidity and bacterial cell counting versus time. Bacteriocine-containing supernatant was prepared by culture centrifugation and subsequent pH adjustment in the range of 6 to 7. Antimicrobial activity of the supernatant was tested on E. coli, S. aureus, A. flavus, and C. albicans using agar diffusion method, determination of total plate count (TPC), and determination of minimum inhibition concentration (MIC). Optimum fermentation was achieved at 20 to 24 hours incubation time. Bacteriocine-containing supernatant showed antimicrobial activity on E. coli only, while in the case of TPC reduced colony count of E. coli and S. aureus was observed due to addition of this supernatant. MIC determination results revealed that MICs of the supernatant on E. coli and S. aureus were 25 and 100% v/v, respectively. Based on overall results, it was concluded that Bacteriocine-containing supernatant prepared from LAB consortium showed antibacterial activity and hence has the potency to be biopreservative.
