Summary:Free radicals are molecular species that have unpaired electron in their outer orbitals. Free radicals can damage important macromolecules such as DNA, proteins, carbohydrates and lipids. Antioxidants are stable molecules that can donate electron to free radicals and inhibit free radical capacity. Antioxidants can be found in various plants, such as pohpohan (Pilea trinervia W.). The objectives of this research were to examine antioxidant activity of pohpohan leaves extracts from three different regions; Bogor, Bandung, and Sukabumi by determining IC50 DPPH and IC50 ABTS; determine total phenolic and flavonoid content; analyze the correlation between total phenolic and flavonoid with IC50 DPPH and IC50 ABTS, and analyze the correlation betwen DPPH and ABTS in the test sample. Extraction was conducted by reflux using increasing polarity solvent. Determination of total phenolic and flavonoid content, IC50 DPPH, IC50 ABTS of each extract were performed by UV-visible spectophotometry. Correlation of total phenolic and flavonoid content with their IC50 of DPPH and IC50 of ABTS and also correlation of two methods were conducted by Pearson’s method. Antioxidant activities of leaves extract pohpohan gave IC50 DPPH in the range of 3.34 –71.33 µg/mL, meanwhile IC50 ABTS 0.48 –3.63 µg/mL. The Highest total phenolic content was given by the ethanol leaves extract of pohpohan Bandung (4.72 ± 0.27 g GAE/ 100 g). The highest total flavonoid content was given by the n-hexane leaves extract of pohpohan Sukabumi (16.94 ± 0.77 g QE/100 g). Total Phenolic content in leaves extract of pohpohan Bogor and Sukabumi had negative and significant correlation with their IC50 DPPH and also with IC50 ABTS of leaves extract of pohpohan Bandung. IC50 DPPH of all sample extract had not significantly positive correlation with their IC50 ABTS. All sample extract of pohpohan were categorized as very strong antioxidant by ABTS method. Phenolic compound were the major contributors in antioxidant activity of leaves of pohpohan Bogor dan Sukabumi by DPPH method. Flavonoid compounds were the major contributors in antioxidant activity of leaves of pohpohan Bandung by DPPH method. DPPH and ABTS methods did not given linear result in antioxidant activity
