The purpose of this study was to evaluate the inhibition activity of canary nut mesocarp and testa extracts on enzymatic activity of tyrosinase, extracted by solvent maceration technique using methanol. Experiments were realized in an optimized condition of the enzyme and L-DOPA substrate concentration at a room incubation temperature. The inhibitory activity was observed by measuring optical absorbance of catalyzed reaction product of L-DOPA by tyrosinase in phosphate buffer at a wavelength of 475 nm. The inhibitory activity was evaluated through tyrosinase activity curve. The 50% inhibiton concentration (IC50) was compared to kojic acid inhibitory activity, showed that IC50 of the canary nut testa, mesocarp extracts and kojic acid solution were respectively 2749.7 ppm, 4661.2 ppm, 121.44 ppm. According to the result, it could be concluded that canary nut testa and mesocarp extract has a tyrosinase inhibitory activity. Compounds that contributed to the inhibition of tyrosinase activity should be flavonoids and polyphenols. The experiments result showed that canary nut testa extract was more potent in inhibiting activity of tyrosinase than mesocarp extract, but both of these extracts were less potent than kojic acid. text
