cosmetic products that many people use in common. However, moisturizing creams can
damage the microflora composition of the skin. The addition of a probiotic bacteria to facial
moisturizing creams is an innovation that is considered useful in balancing the number of
microflora. Therefore, it is considered as an efficient solution to deal with several skin
problems. Lactobacillus plantarum is a probiotic bacteria which is commonly used as raw
material for moisturizing cream. Preliminary research was conducted to determine the
ability of L. plantarum to inhibit the growth of indicator bacteria, Staphylococcus aureus
and Pseudomonas aeruginosa with qualitative Cultural Compatibility Test and quantitative
testing through: (1) Kirby-Bauer method between L. plantarum (108
CFU / mL) and
variations concentration of antibiotic erythromycin against indicator bacteria; and (2)
Microdilution broth to determine the Minimum Inhibitory Concentration (MIC) value,
which is the lowest concentration of L. plantarum cell numbers and variations in the
concentration of erythromycin antibiotics against indicator bacteria using
spectrophotometry. Furthermore, L. plantarum was cultured in a goat milk medium with
inoculum of 10% (v/v), the initial pH of the medium was 6.72 and incubated at 37o
C for 24
hours. The Total Plate Count analysis, pH changes, and total acid production were
condecuted in this study. Based on the results of quantitative and qualitative tests, it shows
that L. plantarum has the ability of inhibiting the growth of indicator bacteria which
produces a zone of inhibition in S. aureus (10 ± 0.44mm) and P. aeruginosa (10 ± 0.33mm)
which is equivalent to the ability of erythromycin concentration 64 ?g / mL. From the MIC
results, it is known that the minimum cell count of L. plantarum in inhibiting S. aureus and
P. aeruginosa is 108
CFU / ml which is equivalent to the ability of erythromycin antibiotics
to inhibit S. aureus and P. aeruginosa each of 4 ?g / mL and 40?g / mL. The results of L.
plantarum growth in goat milk medium had a cell growth rate of 0.22 cells / hour, a change
in pH from 6.72 to 5.18 with a decrease in pH of 0.06 / hour and acid production began at
6 hours with the highest production. total acid 2.25 x 10-3 ppm and the rate of formation
of total organic acids 3.64 ppm / hour. Inhibition test and analysis of the resulting
metabolites have not been carried out. |
