Promethazine HCl is an ethylamino derivative of phenothiazine which is decomposed primarily by oxidation and photolysis. Related substances of promethazine HCl were made by thermal degradation of promethazine HCl in water in the presence of oxygen at 65 C for 48 h. The products were isolated using column chromatography. The purity of isolate was characterized using 2 dimension TLC, ultraviolet-visible and infrared spectrophotometry. The result showed that degradation of promethazine HCl produced 3 pure isolates. The first isolate was assumed to be 10-methylphenothiazine. The second isolate was assumed to be 3H-phenothiazine-3-on with specific C=O groups, while the third isolate was assumed to be promethazine 5-oxide with specific S-O groups. These related substances were assumed to have main similar structure with C=C (aromatic ring), C-N, C-H, and C-S groups, and have conjugated double bond. <br />
