Related substances of promethazine HCl were made by thermal degradation of promethazine HCl in water in the presence of oxygen at 65 C for 48 h. The products were isolated using column chromatography. The purity of isolate was characterized using 2 dimension TLC, ultraviolet-visible and infrared spectrophotometry. The result showed that degradation of promethazine HCl produced 3 pure isolates. The first isolate was assumed to be 10-methylphenothiazine. The second isolate was assumed to be 3H-phenothiazine-3-on with specific C=O groups, while the third isolate was assumed to be promethazine 5-oxide with specific S-O groups. These related substances were assumed to have main similar structure with C=C (aromatic ring), C-N, C-H, and C-S groups, and have conjugated double bond. <br /> text
