This study aims to determine the activity of patchouli leaf oil (Pogostemon cablin B.), ethanol extract, and fraction moringa leaf (Moringa oleifera L.) in inhibiting and killing acne-causing bacteria Propionibacterium acnes and Staphylococcus aureus. In addition, it also determines the class of compounds that act as antibacterial from the selected samples. Patchouli leaf oil is obtained through a process of steam distillation. Extraction was carried out by maceration method using 96% ethanol, then fractionated by liquid-liquid extraction method using n-hexane, ethyl-acetate, and ethanol-water. The antibacterial activity test was carried out qualitatively using the agar disc diffusion method and TLC-bioautography, as well as quantitatively by determining the value of the minimum inhibitory concentration (MIC) using the microdilution method and the minimum bactericidal concentration (MBC) using the agar diffusion method. The ethanol extract, n-hexane fraction, and ethyl acetate fraction of moringa leaves did not show antibacterial activity in Propionibacterium acnes ATCC 11827 and Staphylococcus aureus ATCC 6538. Patchouli leaf oil has strong antibacterial activity against Propionibacterium acnes ATCC 11827 and Staphylococcus aureus ATCC 6538 with MIC–MBC values of 312.5 µg/mL and 156.25–312.5 µg/mL. Compounds that have antibacterial activity are thought to be sesquiterpenes. text |
