In this study, in vitro shoot culture in an ebb-and-flow bioreactor was the preferred method for better propagation compared to conventional methods. The objective of this research was to compare the growth performance and yield of these sweetening compounds in S. rebaudiana under conditions without elicitation (control) and with 10 µM methyl jasmonate (Me-J) elicitation treatment. Growth performance was determined based on parameters such as the increase in the number of shoots, biomass increase, and relative growth rate. Elicitation using 10 µM Me-J showed improved growth performance based on measurable and calculated parameters. The increase in the number of shoots in the control and treatment groups was 2.8 and 2.9 times, respectively. The increase in biomass in the control and treatment groups was 13 and 13.7 times, respectively. The relative growth rate in the control and treatment groups was 0.184 and 0.189 mg.mg-1.day-1. The average stevioside content in the leaves of S. rebaudiana in the control and treatment groups was 407.94 and 655.21 µg.g-1 dry weight (DW) of leaves, respectively. The average Reb-A content in the leaves of S. rebaudiana in the control and treatment groups was 437.15 and 211.63 µg.g-1 DW of leaves, respectively. Elicitation with methyl jasmonate increased the stevioside content in S. rebaudiana by 60.6%, but it decreased the Reb-A content by 51.6%. Overall, steviol glycosides increased in the treatment, but it is suspected that the time required for Me-J to support the synthesis of Reb-A from stevioside was not achieved, resulting in stevioside being the predominant steviol glycoside observed in the extract text |
