The addition of phosphorylated cilembu sweet potato polysaccharide shows the ability as a stabilizer and can increase the antioxidant activity in this nanoemulsion system. This study aims to optimize the composition of nanoemulsion combination of basil leaves extract and cilembu sweet potato polysaccharide using Box-Behnken experimental design. Extraction of basil leaves was carried out by UAE (Ultrasound-Assisted Extraction) method using 60% ethanol solvent and 1% citric acid solution in the ratio of 60:40). The nanoemulsion formula components included Tween 20, sunflower seed oil, hexylenglycol, glycerin, basil leaves extract, cilembu sweet potato polysaccharide and arginine. Screening of formula parameters was conducted using the OVAT (One Variable at Time) method. Box-Behnken experimental design was conducted using three factors identified from the preliminary analysis. Then the optimum formula obtained was characterized and tested for physical stability for 28 days. The extraction results of basil leaves obtained an extract yield of 6.68% and the extraction results of cilembu sweet potato polysaccharides amounted to 7.06%. The optimum formula composition required an oil-extract ratio (2.2% : 1%), a hexylenglycol concentration of 1%, and a cilembu sweet potato polysaccharide concentration of 2%. The optimum formula had a globule size of 195.10 nm. Antioxidant activity test between extract and formula using DPPH method increased by 12.42%. The zeta potential value of the optimum formula was -42.57 mV. The optimization results of nanoemulsion formula combination of basil leaves extract and cilembu sweet potato polysaccharide using Box-Behnken experimental design successfully met the target response of globule size below 200 nm so that it showed great potential for further development as an antioxidant solution. text
