Rifampicin is unstable in solution which becomes a problem in analysis with chromatography or spectrophotometry. This study aimed to develop a fast, inexpensive, simple, and reliable method for the determination of rifampicin (RIF), isoniazid (INH), pyrazinamide (PZA) dan ethambutol hydrochloride (ETH) based on infrared spectrophotometry in combination with multivariate calibrations, especially partial least square (PLS). The utility of chemometrics-assisted infrared spectrophotometry based on PLS for determination of drug combinations has been developed. Analysis of active component using infrared spectrophotometry-PLS is carry out in three phases, namely calibration, validation and sample assay. Calibration model is evaluated by the value of root mean square error of calibration (RMSEC), root mean square error of cross validation (RMSECV), root mean square error of prediction (RMSEP) and predicted error sum of square (PRESS) as precision criteria and coefficient of determination (R2) as accuracy criteria. Results of the study showed that multivariate calibration-assisted infrared spectrophotometry based on PLS can be used for determination of rifampicin, isoniazid, pyrazinamide dan ethambutol hydrochloride. The optimized wavenumber range for determination of RIF contents was 433 � 1552 cm-1 and 1756 � 3412 cm-1, 433 � 873 cm-1 for INH, 1714 � 2756 cm-1 for PZA, and 1552 � 2970 cm-1 for ETH. The PRESS values of RIF, INH, PZA and ETH contents was 0,00392%, 0,011724%, 0,007604% and 0,003368% respectively. RMSEP values of RIF, INH, PZA and ETH contents was 0,01979%, 0,03424%, 0,027575% and 0,018351% respectively. The value of R2 was 0,990, 0,977, 0,992 and 0,998 for RIF, INH, PZA and ETH respectively.
