Thus it is very potential to cultivate as a new agrochemical product.The research aim is to determine amino acid qualitatively and quantitatively in black cucumber (Holothuria edulis). The identification and determination of amino acid was carried out by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). TLC operational condition was as follows silica gel plat 60 F 254 was used as stationary phase and two types of mobile phase were buthanol : acetic acid : water (4 : 1 : 1) and chloroform : methanol : water (2 : 2 : 1). The chromatogram showed 7 amino acid, arginine, histidine, leusine, lisine, methionine, phenylalanine and valine. Analysis by HPLC was carried out using buffer A (mixture of sodium acetate 0,025 M (pH = 6,5); NaEDTA 9%(v/v); methanol 1%(v/v); THF as mobile phase; and reagent B (methanol 95%(v/v) as mobile phase, ultra Techspere-ODC was used as stationary phase, and pre-column derivation detection system. The result demonstrates that black cucumber (Holothuria edulis) consists of 17,41% non-essential amino acid and 9,56% essential amino acid. Composition of non-essential amino acid are 3,20% aspartic acid; 4,25% glutamic acid; 1,39% serine; 2,73% glisine; 2,81% arginine; 0,90% tyrosine; 2,13% alanine. And composition of essential amino acid are 0,33% histidine; 1,41% threonine; 0,71% methionine; 1,08% valine; 1,03% phenylalanine; 0,80% isoleusine; 1,71% leusine; and 2,49% lysine. text |
