antioxidant. High performance liquid chromatography (HPLC), UV Vis spectrophotometry, and TLC <br />
<br />
densitometry have been used for determination of epicatechin in a variety of samples. The <br />
<br />
purpose of this study was to obtain and validate the method of determining epicatechin contents <br />
<br />
in white tea extract using TLC densitometry and determination of epicatechin contents in sample. <br />
<br />
The first step of this study was characterization of crude drug and extract. Further, optimization of <br />
<br />
mobile phase and saturation of chamber were done to obtain optimum condition. The validation <br />
<br />
of methods consist of evaluations on specificity, linearity, limit of detection, limit of <br />
<br />
quantification, accuracy, and presision. Contents of epicatechin were also determined in white tea <br />
<br />
extract sample using validated methods. The best composition of mobile phase were chloroform– <br />
<br />
aseton–formic acid (5:4:1) with 30 minutes for chamber saturation. The values of linearity, <br />
<br />
accuracy recovery value, intraday precision, interday precision, limit of detection, and limit of <br />
<br />
quantification were r = 0.999, r <br />

= 0.998; 90.01%-92.65%; 2.001% and 1.973%; 2.011% and <br />
<br />
1.934%; 0.537 µg/ spot; 1.628 µg/spot respectively. Based on the methods validation results, <br />
<br />
epicatechin contents in white tea extract sample can be determined using TLC densitometry. The <br />
<br />
average value of epicatechin contents in white tea extract sample was 1398.11 ± 24.61 µg/ mL. <br />
