One of tea components is epicatechin, which has biological effect as strong

antioxidant. High performance liquid chromatography (HPLC), UV Vis spectrophotometry, and TLC

densitometry have been used for determination of epicatechin in a variety of samples. The

purpose of this study was to obtain and validate the method of determining epicatechin contents

in white tea extract using TLC densitometry and determination of epicatechin contents in sample.

The first step of this study was characterization of crude drug and extract. Further, optimization of

mobile phase and saturation of chamber were done to obtain optimum condition. The validation

of methods consist of evaluations on specificity, linearity, limit of detection, limit of

quantification, accuracy, and presision. Contents of epicatechin were also determined in white tea

extract sample using validated methods. The best composition of mobile phase were chloroform–

aseton–formic acid (5:4:1) with 30 minutes for chamber saturation. The values of linearity,

accuracy recovery value, intraday precision, interday precision, limit of detection, and limit of

quantification were r = 0.999, r

2

= 0.998; 90.01%-92.65%; 2.001% and 1.973%; 2.011% and

1.934%; 0.537 µg/ spot; 1.628 µg/spot respectively. Based on the methods validation results,

epicatechin contents in white tea extract sample can be determined using TLC densitometry. The

average value of epicatechin contents in white tea extract sample was 1398.11 ± 24.61 µg/ mL.
text
