function disorders, cell structure damage, degenaratif disease and cancer. Most plants may have
antioxidant activity when it contains compounds that can scavange free radicals, such as
flavonoids and phenols. Black and white sesame seed that usually used as a food supplement is
thought to have antioxidant activity. This research aims to test the antioxidant activity of black
and white sesame seed through the determination of IC50 DPPH (2,2-diphenyl-1-picrylhydrazyl),
IC50 ABTS (2,2-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid ) and EC50 FRAP (ferric reducing
antioxidant power), determining the total phenol and flavonoids, correlations of total phenolic
and flavonoid content in each extract with their IC50 of DPPH, IC50 of ABTS, EC50 of FRAP and
correlation of three antioxidant methods in sample extracts. Determination of IC50 of DPPH, IC50 of
ABTS, EC50 of FRAP, total phenolic, and flavonoid content were performed by UV-visible
spectrophotometry and its correlation IC50 of DPPH, IC50 of ABTS, EC50 of FRAP were analyzed by
Pearson????s method. Antioxidant activity assays of black and white sesame seed showed that nhexane black sesame seed extract gave the lowest IC50 of DPPH (8,879 µg/mL), ethanol black
sesame seed extract gave the lowest IC50 of ABTS (22.907 µg/mL) and ethanol black sesame seed
extract gave the lowest EC50 of FRAP (222.401 µg/mL). Ethanol black sesame seed extract gave the
higest phenolic content (1.569 g GAE/100 g) and ethyl acetate white sesame seed extract gave the
higest flavonoid content (4.290 g QE/100 g). ) Total phenol content in black sesame seed extract
had negative and significant correlation with their IC50 of ABTS and EC50 of FRAP, while flavonoid
content in white sesame seed extract had negative and significant correlation with their EC50 of
FRAP. The results of correlation test between DPPH, ABTS and FRAP methods showed that white
sesame seed extract there was positive and significant correlation between IC50 DPPH and EC50
FRAP, while black sesame seed extract gave positive and significant correlation for IC50 ABTS and
EC50 FRAP. All seed extracts of black and white sesame were categorized as very strong
antioxidant by DPPH methods. Phenolic compounds were major contributor in antioxidant activity
of black sesame seed extract by ABTS and FRAP methods, while flavonoid compounds were major
contributor in antioxidant activity of white sesame seed extract by FRAP method. DPPH and FRAP
methods gave linear results in antioxidant activity of white sesame seed extracts, while ABTS and
FRAP methods showed linear result in black sesame seed extract.
