Summary:Free radicals are atoms or groups of atoms that have one or more unpaired electrons. Free radicals can against and bound with electron of the other molecule. Negative effect of free radical can be inhibited by antioxidant. Antioxidants are compound that can inhibit oxidation reaction by scavenging free radical. Chayote had been proved as having antioxidant activity. The objectives of this research were to determine the antioxidant activities of fruit chayote (Sechium edule) by determining IC50 of DPPH scavenging activity and EC50 of phosphomolybdenum capacity, total phenolic, flavonoid and carotenoid content, their correlation with the IC50 DPPH and EC50 of phosphomolybdenum, and correlation between IC50 DPPH and EC50 of phosphomolybdenum. Extraction was performed by reflux apparatus using three solvents with increasing polarity. Determination of IC50 of DPPH, EC50 of phosphomolybdenum, total phenolic, flavonoid and carotenoid content of each extract were carried out by UV-visible spectrophotometry and its correlation with IC50 of DPPH and EC50 ?» ????í?????????í????????¢­±??????????±????± •?•?????????±­ ¢????±•?????????????????? method. The highest antioxidant activity by DPPH method (IC50 9.32 µg/mL) was given by nhexane fruit extract of chayote from Lembang, while using phosphomolybdenum method was showed by ethyl acetate fruit extract of chayote from Semarang (EC50 209.87 µg/mL). Total flavonoid content of all fruit extracts had significantly negative correlation with IC50 DPPH and EC50 phoshomolybdenum value. Flavonoid compounds in chayote fruit extracts from Semarang, Malang and Lembang were the major contributor in their antioxidant activity by DPPH and phosphomolybdenum assays. DPPH and phosphomolybdenum methods gave linear results in all of sample extracts.
